Retatrutide Research: FDA Status, Structure and Receptor Pharmacology
Retatrutide, known in the literature by its development code LY3437943, is Eli Lilly’s investigational triple agonist: a single peptide that activates the GIP, GLP-1 and glucagon receptors. This page is a research reference for US readers. It covers where the molecule stands with the FDA, how its design builds on tirzepatide, what the published receptor pharmacology says, and what a laboratory should expect to see on a certificate of analysis for a peptide this complex.

Is retatrutide FDA approved?
No. As of September 2026 retatrutide is an investigational drug in the United States. No application has been filed and nothing has been approved. In July 2026 Lilly said it plans to submit a Biologics License Application (BLA) to the FDA in the first quarter of 2027. FDA review follows submission, so no approval date can be given yet.
Research-grade material is a separate matter. A peptide sold as a laboratory reference material is not an FDA-approved drug, is not made under drug-manufacturing (cGMP) controls, carries no sterility or endotoxin assurance, and has no drug labeling. It is for in vitro laboratory work only.
From two receptors to three
Retatrutide comes from the same Lilly program that produced tirzepatide, and the two molecules are close relatives:

The design moves are small but deliberate. Aib at position 2 blocks the enzyme DPP-4, which inactivates native incretins by clipping their first two residues. The α-methyl-leucine at 13 and Aib at 20 tune activity across the three receptors, and moving the fatty diacid from lysine 20 to lysine 17 changes where the albumin-binding group sits on the helix. Receptor pharmacology in numbers
All three receptors are class B G-protein-coupled receptors that signal through Gs and cyclic AMP, so potency is measured as cAMP accumulation in cells expressing one human receptor at a time. Against each receptor’s own hormone, Coskun et al. reported:

So, like tirzepatide, retatrutide leans toward the GIP receptor. Two cautions when using figures like these. They are functional potencies (EC50), not binding affinities, and the two are not interchangeable. And for lipidated peptides, the amount of albumin or serum in the assay shifts apparent potency, because the diacid binds albumin and lowers the free concentration. Figures from different assay conditions should not be compared directly.
In animals, the discovery work used GLP-1 receptor-knockout and GIP receptor-knockout mice to separate each receptor’s contribution, which is the standard way to take apart a multi-receptor response.
What a certificate of analysis should show
For a 39-residue lipidated peptide, a single HPLC purity figure is not enough on its own. A useful certificate shows:
- Intact mass by mass spectrometry, matching about 4,731 g/mol. This is the identity check. It also separates retatrutide from tirzepatide, which differs by about 80 g/mol and behaves similarly on a column.
- HPLC purity with a readable chromatogram. The C20 diacid dominates retention, so impurities that keep the lipid can sit close to the main peak.
- Peptide content, not just purity. A vial’s weight includes counter-ions and water, so the peptide content is below the nominal fill. Our note on amino acid analysis and net peptide content explains how it is measured.
