CJC-1295 and Ipamorelin USA

CJC-1295 and ipamorelin get ordered together so routinely that the pairing is often treated as one product. It is not. One is a 29-residue GHRH analog, the other is a five-residue secretagogue with two D-amino acids and a non-standard residue, and they act at different receptors, age differently in solution and need to be confirmed separately on a certificate. This page explains the two molecules, when the blend is the right choice and when it is not, and how to order either within the United States.

Two different molecules

CJC-1295 without DAC, also written modified GRF(1-29), is the first 29 residues of GHRH with four substitutions: D-Ala at position 2, Gln at 8, Ala at 15 and Leu at 27. Mass is close to 3,368 Da. The DAC version adds a maleimidopropionyl linker of roughly 300 Da and is a different compound with different behaviour; everything we stock is the no-DAC form. Ipamorelin is Aib-His-D-2-Nal-D-Phe-Lys-NH2, a pentapeptide of about 712 Da with an aminoisobutyric acid at the N-terminus, two D-residues and a C-terminal amide. It belongs to the growth hormone secretagogue family acting at GHSR-1a. Blend or two vials

We supply both forms:

• CJC-1295 and ipamorelin blend 10 mg: 5 mg of each, co-lyophilised.
• CJC-1295 no DAC 5 mg alone.
• Ipamorelin 5 mg and 10 mg alone.

The blend fixes the mass ratio at 1:1, which at 3,368 Da against 712 Da is a molar ratio of roughly 1:4.7 in favour of ipamorelin. That is fine where the protocol is written around it. It is not fine where each compound needs its own control arm, where the molar ratio is a variable, or where the two will be used on different days, because the CJC-1295 component sets the shelf life of the whole solution. In those cases two vials are the better purchase even though the blend is convenient.

Reading a blend certificate

A blend chromatogram should show two well-separated main peaks, since a 29-mer and a pentapeptide elute far apart on a reversed-phase gradient, and each should be assigned by mass: about 3,368 and about 712 Da. The figure to be wary of is a single purity percentage for the whole vial with no breakdown. That number describes the chromatogram, not the composition, and it cannot tell you whether the vial holds 5 mg of each or 8 mg of one and 2 mg of the other. Why the two age differently

CJC-1295 carries a methionine at position 27 and several asparagines; the methionine oxidises in the presence of dissolved oxygen and the asparagines deamidate, so reconstituted solution should be kept cold, aliquoted and used within days. Ipamorelin has neither residue, has two D-amino acids that resist proteolysis, and is markedly more robust. Both are stable as lyophilised powder at minus 20 °C in a sealed vial.
