You Cannot Put a Purity Figure on a Fraction

Ask for thymalin’s sequence, its formula or its molecular weight and there is no answer to give. That is not a documentation gap. Thymalin is a preparation rather than a compound, and working out what follows from that fact is more useful than any specification sheet.

What “fraction” actually means here

The literature describes thymalin as a polypeptide fraction recovered from thymic tissue. A fraction is whatever a separation step retained after discarding the rest, which makes the procedure – not a chemical formula – the thing that defines it.

The consequence is a population of polypeptides rather than one molecule. Composition tracks the source tissue and the particular extraction and purification steps used on it, so there is no single sequence, no single formula and no single mass to report.

Thymalin is not thymosin alpha-1, and neither is thymosin beta-4

Catalog listings place these near one another, and thymalin is occasionally presented as a synonym for thymosin alpha-1 or as its crude form. That is wrong on both counts.

Thymosin alpha-1 is a defined, acetylated 28-residue peptide with a calculated mass close to 3108 daltons and its own CAS registry number. It can be synthesized and characterized the way any peptide is. Thymalin cannot, because a preparation is not a molecule – even when that preparation happens to contain molecules that are separately available in defined form.

Thymosin beta-4 is a third and distinct entity, a 43-residue actin-binding peptide whose sequence bears no relation to thymosin alpha-1. The shared word thymosin points at tissue of origin, not at a structural family.

Three of the four certificate fields do not exist

A conventional certificate for a defined peptide gives a sequence, a calculated mass, a measured mass and a purity value, each with a single clear meaning. Applied to a fraction, most of those fields have nothing to hold.

  • Sequence. Undefined, by construction.
  • Mass. A mass spectrum of a fraction yields a distribution rather than a peak. That distribution can be characterized, but it cannot be checked against a calculated value, because no calculated value exists.
  • Purity. An area percentage expresses one chromatographic peak as a share of the total. A fraction is deliberately many peaks, so a high area percentage would be evidence that the material is something other than what it claims to be.
  • Identity. Where it is established at all, it is established by matching a reference profile, not by matching a number.

Plenty remains that a document for a fraction can legitimately report: the source material, the process, total protein or total nitrogen content, a chromatographic or electrophoretic profile, and contaminant testing. Those are genuine measurements – just a different set of them.

Lot-to-lot variation is built in

Two lots of a synthetic peptide carrying the same sequence are the same compound, and whatever separates them is impurity. Two correctly produced lots of a tissue-derived fraction can differ in composition simply because the starting tissue did.

Profile comparison, not a numerical specification, is therefore the appropriate control. It also means the phrase batch-to-batch consistency carries a different meaning for a fraction than it does for a synthetic molecule.

How to read the paperwork

When a certificate for a fraction reports one mass and one purity percentage, a synthetic-peptide template has been filled in with figures that do not describe the material in the vial. That is usually the wrong form rather than an attempt to mislead, but the effect on a reader is the same.

The informative alternative states what the material is, what process made it, and what was actually measured. Being able to tell those two documents apart on sight is the practical payoff of understanding the distinction.

Thymalin is not alone in this category

A number of items shelved beside defined peptides are themselves fractions or mixtures. Cerebrolysin, for instance, is described as a peptide preparation obtained from porcine brain tissue, and a range of organ-derived extracts are described along the same lines.

A single question sorts them. Does the compound have one sequence and one calculated mass? If so, it is a defined molecule and a conventional certificate suits it. If not, it is a preparation and requires documentation built for a mixture. Neither category ranks above the other; they are different kinds of thing, characterized in different ways.

The contents of an appropriate document

Source and process stated plainly; a chromatographic or electrophoretic profile together with a reference profile to compare it against; a total content figure obtained by a method suited to mixtures; and contaminant testing. A calculated molecular mass printed on such a document indicates that the template rather than the material dictated what got written down.

The defined peptides that share the name

Characterized in the conventional way, thymosin alpha-1 is a defined 28-residue sequence; see thymosin alpha-1 sequence, mass and detection. The separate and unrelated thymosin beta-4 is treated in thymosin beta-4 versus the TB-500 fragment.
