A 0.98 Dalton Shift Is Your Peptide Ageing
An amide becomes an acid, and the mass barely moves
Asparagine and glutamine each carry an amide group out on the side chain. Deamidation turns that amide into a carboxylic acid: an NH2 leaves, an OH takes its place, and the nominal mass changes by 0.984 Da. That is the entire origin of the number. It also explains why the resulting species parks itself immediately above the parent on a spectrum rather than somewhere obvious further along, and why an instrument without much resolving power will pass over it without comment.
Chemically the transformation goes by way of a cyclic succinimide, and the ring that forms has two ways to open again. Take one and you get the aspartate anyone would predict. Take the other and you get isoaspartate, where the backbone now runs through what had been the side chain. Since the two products weigh identically, no mass spectrum by itself can say which of them occupies the vial.
The residue next door sets the pace
Susceptibility is not a property of a peptide considered as a whole. It is a property of one small neighborhood, governed by whichever residue follows the asparagine, because that neighbor decides how easily the succinimide ring can close. Glycine has no side chain to get in the way, which makes Asn-Gly the canonical fast case, degrading far more readily than Asn-Val or Asn-Ile, where a bulky neighboring side chain obstructs the closure.
Follow that through and two peptides sitting in identical storage can behave nothing alike. A stability observation made on one sequence travels badly to the next.
What pushes the reaction along
| Variable | Effect |
| Moisture | Water is a reactant here, which is the concrete reason freeze-dried material is kept dry and the reason residual water content matters for more than what it does to the mass on the label |
| Warmth | Raises the rate, in the ordinary way |
| pH | A strong dependence, with neutral and alkaline conditions favoring succinimide formation |
Put those three together and the practical implication is that the clock on this particular degradation route begins at reconstitution, not at dispatch. Routes that run in parallel with it are covered in light, oxygen and temperature, and limiting exposure once material has gone into solution is the subject of aliquoting and vial entry.
Finding it on a report
On the mass spectrum, look for a satellite roughly 0.98 Da higher than the parent signal. Two conditions have to hold before that is even possible: the instrument must resolve a gap that narrow, and the report must reproduce enough of the spectrum for satellites to be visible. One tightly cropped label around the main peak will hide the whole question.
On the chromatogram, the evidence takes a different form. Stripping an amide makes the molecule more acidic, and that change of character alters its behavior on a reversed-phase column, so the degraded form frequently turns up as a shoulder or as a properly resolved peak eluting later.
Whether any of that counts against the purity figure depends on detection wavelength. At 214 nm the detector is watching the peptide bond, so deamidated material is counted as impurity. At 280 nm it may escape counting, according to what the sequence contains. A shoulder’s meaning is unpacked in reading an HPLC chromatogram, and the other species a chromatogram silently tallies are listed in deletion, truncation and oxidation impurities.
What the satellite does not tell you
Its presence establishes that a chemical change happened. It carries no date. It cannot separate an artifact of synthesis from an artifact of storage. It attaches no quantity to the affected fraction unless the chromatogram has actually been integrated with that in mind. And it bears in no way on suitability for use in a person or an animal, which sits outside the reach of any identity or purity test.
All of the above concerns laboratory research material and the published literature. Nothing supplied is a drug or a supplement, and none of it is intended for administration to people or to animals.
