Three Tests US Buyers Routinely Treat as One

Three terms get swapped around as though they were synonyms, and they measure three different things. The distinctions are worth holding on to, because material can pass one and fail another — and because none of the three appear on a routine identity and purity certificate.

Bioburden

Bioburden is a count: how many viable microorganisms are present in or on a material, established by culturing a sample and counting what grows. A quantitative measure of contamination, not a pass-or-fail state.

Its limitation is that it counts what will grow under the conditions chosen. Organisms with different nutritional requirements, different temperature optima, or sitting in a viable but non-culturable state go uncounted. A bioburden number describes what that method could detect.

Sterility

Sterility is the absence of viable organisms, tested by incubating material in growth media and watching for growth. The outcome is binary rather than a count.

The statistical structure of sterility testing is widely misread. The test runs on a sample of units, not on every unit, so a pass establishes that nothing grew in the units tested. Where low-level contamination is sparsely distributed across a batch, a sampling plan can pass while contaminated units remain in it. Sterility is therefore assured primarily by a validated process, with the test acting as confirmation rather than as the assurance itself.

Pyrogens and endotoxin

A pyrogen is anything that produces a fever response. The most significant class in practice is bacterial endotoxin — lipopolysaccharide from the outer membrane of Gram-negative bacteria.

The property that matters is that endotoxin is not alive. It is a molecule; it survives autoclaving, and it stays active long after the organism that made it has been destroyed. A preparation can therefore be sterile and pyrogenic simultaneously, which is the single most common misunderstanding in this area.

Endotoxin is measured by the limulus amoebocyte lysate assay or by recombinant factor C, both of which detect the molecule rather than the organism. Endotoxin, sterility and the limits of research grade covers what research-grade material does and does not carry.

Why none of them show up on an identity certificate

HPLC and mass spectrometry measure chemical composition. They are blind to microbiological content: a chromatogram looks the same whether the sample is sterile or heavily contaminated, because bacteria and endotoxin produce no peaks at peptide wavelengths.

These are separate tests needing separate samples, separate methods and separate facilities, and they are not part of routine peptide characterization. A certificate that does not report them is not implying a result — it means those tests were not run. What a third-party peptide test actually measures covers the scope of the usual document.

All material is supplied for laboratory research use only. It is not a drug, not a supplement, and not for use in humans or animals.
