What a Warm Package Means for the Vial Inside
Vials leave the building at ambient temperature while the label asks for cold storage, and that contradiction is usually the first thing a new buyer queries. Two facts resolve it: dry powder and dissolved peptide tolerate warmth on completely different scales, and a courier leg is counted in days.
Why an average temperature tells you almost nothing
Degradation climbs exponentially as temperature rises, so totaling a transit profile and dividing by its length discards the information that matters. An hour spent at 40 degrees costs more than an hour at 20 gives back.
Mean kinetic temperature is the arithmetic that survives this. Defined as the single temperature that would inflict the same cumulative degradation as the real fluctuating profile, it sits above the plain arithmetic mean without exception, and the distance between the two widens as excursions sharpen. Consider a shipment averaging 22 degrees that touched 45 in short bursts: its mean kinetic temperature lands well above 22.
Framed that way, one brief severe spike outranks a long mild warm spell.
Chemistry that stalls without water
Water is the shared requirement behind most peptide degradation. Hydrolysis consumes it outright; deamidation proceeds through a hydrated intermediate; disulfide exchange depends on a molecular mobility that a dry solid withholds. Strip the water out and reaction rates drop by orders of magnitude.
Hence a cake lyophilized to low residual moisture shrugs off warmth that would measurably harm the same peptide in solution. Changes a solution registers within hours take days at ambient temperature in the dry state. Depth on the solid itself sits in the appearance of a freeze-dried cake, while the assay that puts a number on dryness appears under residual water by titration.
One qualification belongs here. Low residual moisture describes a particular lot, not lyophilized material as a category. Where a cake has collapsed or come off the shelf under-dried, it holds more water and carries correspondingly less margin in transit.
Conditions that do the real damage
Brief warmth against a sealed, dry vial rarely amounts to anything. Three other situations earn the attention instead.
| Situation | Why it matters |
|---|---|
| Heat acting on a hydrated solid | A compromised seal admits humidity, after which warmth works on wet material. Damage comes from the pairing, not temperature alone. Seal integrity is treated in crimping and headspace. |
| Arrival frozen, opened straight away | Cold solid pulls condensation out of room air. Consequences for a mass reading are covered in gravimetry on hygroscopic solids. |
| Exposure that drags on | A parcel held for weeks somewhere warm is a wholly different proposition from a two-day transit. |
First moves once the parcel lands
Bring the vial to room temperature before the seal is broken, examine the cake and the closure, and record the arrival condition before anything goes away. The full sequence appears in inbound inspection steps; conditions for the period afterwards are set out in keeping lyophilized peptides stable.
