Storing Lyophilised Peptide So the Certificate Still Applies

Take a vial straight out of the refrigerator, break the seal, and room air meets a surface sitting below its dew point. Moisture condenses onto the powder immediately. Lyophilized peptide is hygroscopic, so that water is not going anywhere once the vial is closed again, and what it enables is precisely the hydrolysis freeze-drying was performed to prevent. Of all the avoidable losses in peptide handling, this is the commonest.

The remedy is procedure, not equipment

Let the sealed vial reach room temperature before the cap comes off. Twenty to thirty minutes covers it, depending on how large the vial is. Handle the powder somewhere dry if the option exists, reseal without dawdling, and keep a vial that gets opened repeatedly under desiccant.

Each reopening repeats the exposure

Removing a vial, warming it, opening it and putting it back means crossing the dew point once more, and moisture gets a fresh opportunity every time. Aliquoting is the standard answer: divide the material into single-use portions at the first opening so that bulk stock is disturbed exactly once rather than over and over. Keep a count of how many times any working aliquot has been opened.

What the dryness is buying

Freeze-drying strips out the water that most degradation chemistry depends on. Hydrolysis of the peptide bond, deamidation at asparagine and glutamine, oxidation of methionine, cysteine and tryptophan: all of them run far more slowly through a dry solid than through aqueous solution. Held properly, lyophilized material can last years. The identical peptide sitting in solution at room temperature may show measurable degradation inside days.

Temperature, and why nothing ships cold

Refrigerated storage at 2–8 °C is the standing instruction. Dry powder generally tolerates a short ambient transit, which is the reason peptides move without a cold chain, but tolerating transit is not the same as tolerating storage. Degradation rates climb steeply with temperature, and a vial left at room temperature for weeks has an unknown purity no matter what its original analysis reported.

Light and oxygen

Tryptophan, tyrosine and phenylalanine make a sequence photosensitive; methionine and cysteine make it prone to oxidation. Amber vials, opaque secondary containers and short bench exposure all cut the exposure down. Where a sequence is particularly oxidation-sensitive, purging the headspace with an inert gas before resealing is standard practice.

Once it is in solution

Reconstitution costs a substantial amount of stability, and what remains depends on both sequence and buffer. Refrigerate solutions, keep them briefly, and make them fresh wherever the experiment permits. pH is part of the picture too: many peptides are least stable at the extremes, and sequences containing aspartate are especially liable to isomerization under acidic conditions. Any solution that must be kept should be split into aliquots rather than held as one stock and sampled repeatedly.

Finding out whether it happened

Very little of this is visible, which is what makes it dangerous; a partly degraded sample looks exactly like an intact one. Inspection by eye catches gross failures only. A cake that has collapsed, discolored or gone sticky points to moisture ingress, while cloudiness, visible particulate or a refusal to dissolve fully points to aggregation. Neither is a dependable early warning. What settles the question is re-running analytical HPLC on the stored material and laying the trace alongside the original chromatogram.
