Two analogs of one 44-residue parent

Native GHRH is a 44-residue peptide, and the two compounds on this page are synthetic analogs that depart from it in opposite directions. Sermorelin shortens it: the chain stops at residue 29 and ends in a C-terminal amide, which is why it is written GHRH(1-29)-NH2. The chain opens with Tyr-Ala, nothing is attached to it, and the amide form weighs about one dalton less than the free acid. Tesamorelin keeps all 44 residues and instead carries a trans-3-hexenoyl group on the N-terminus, so its calculated mass must include that acyl group as well as the chain.

The report we would expect for a GHRH analog

At the moment there is no published third-party report for sermorelin or tesamorelin on our site. Tested lots are added to the certificates page as reports arrive, and pointing at a report for some other compound would be worse than saying so. These are the items we would want on any vendor’s certificate for either peptide.

• A named laboratory and a method section. Without the name the figures cannot be queried, and without column, gradient and detection wavelength an area percentage on a 29- or 44-residue chain means little.
• A mass check against the right molecule. For sermorelin the calculated mass must be the amidated form, not the free acid; the two are about one dalton apart and elute close together, so HPLC alone cannot separate them. For tesamorelin it has to include the hexenoyl group, or the check was run against the wrong structure.
• An eye on oxidation. Both sequences carry a methionine residue, and an oxidized chain appears as a satellite 16 Da above the parent mass. A report that prints the observed mass but says nothing about that region has not looked.
• Aggregation state for the 44-mer. Size-exclusion chromatography or dynamic light scattering shows whether a long chain is present as single molecules or as clusters.
• Net peptide content on its own line. Label milligrams are gross weight; after counter-ion (usually TFA) and residual water, net content across our catalog is typically 70–90% of label, a measurement covered in net peptide content explained.
• Batch photographs. Our vials carry no printed lot number, so cap color and crimp color checked against the photographs are the only link between a vial and a report.

4. How is the vial you receive tied to the certificate online: a printed lot number, or batch photographs?
